Collagen is a protein in mammals providing support and structural organization to cells in most tissue types. It
contains a high concentration of the 4-Hydroxy-L-proline, so 4-Hydroxy-L-proline has been historically used as an
estimate of collagen content. A new simple procedure for the hydrolysis of collagen in natural products and materials
and determination of collagen through a hydrolyzed product 4-Hydroxy-L-proline were developed. Factors to be
investigated including the volume of hydrochloric acid solution, hydrolysis time, and hydrolysis temperature. Optimal
results are obtained consisting of 1mL of HCl (6N) for 2 mg of collagen at 110 °C with short hydrolysis time (6 hours).
It has a high efficiency of 96.7 % and good repeatability with 6 hydrolyzed independent samples in the same condition
and RSD% under 5 %.
