Microsomes isolated from rat liver containing enzymes for both phase I and phase II of drug metabolism have crucial application in drug development such as evaluating drug metabolism and drug influence on CYP450s. In order to determine the optimal age of rats and proper liver preservation condition to obtain highest yield of microsomes, the effect of rat’s age and the duration/temperture of liver preservation on the yield of microsomes was investegated. The result indicates that seven-week old rats yielded the highest obtained microsomes among rats from the age of two weeks to eight weeks. The research result also shows that the yield of microsomes was not influenced by the storage of liver at -15°C or -80°C within six months but significantly decreased in the case of 4°C.
