An HPLC technique with PDA detector has been developed for simultaneous quantification of four lignans including phyllanthin (PLT), hypophyllanthin (HPL), niranthin (NRT) and nirtetralin (NTT) in Phyllanthus amarus Schum. et Thonn. plant samples. Chromatographic separation was achieved on a Gemini NX C18 column (150 x 4.6 mm, 5 mm) with an isocratic mobile phase consiting of methanol – 0.1 % phosphoric acid (56 : 44, v/v). The method was successfully validated for system suitability, specificity, linearity range, precision (including repeatability and imtermediate precision) and accuracy. The method was applied for determination of lignans in P. amarus plant samples or dry extract and tablets containing P. amarus.
