Summary
The CD4 T-lymphocyte count test plays an important role in the diagnosis and monitoring response to treatment of diseases related to the immune system, especially HIV infection. The purpose of this study was to establish the internal quality control (IQC) samples for enumerating CD4 T-lymphocytes. The stability of T-CD4 cells in whole blood were optimized by using the preservative solution (Streck®) at different dilution ratios. The CD4 T-lymphocyte counts were determined using BD FACT Canto II flow cytometer. The optimal ratio of preservative solution and whole blood was applied to develop the IQC panels. Each sample panel contained 2 blood samples with normal and low CD4 T-lymphocyte concentration. The stability and homogeneity of IQC panels were evaluated. The results showed that the samples in IQC panels were homogenous, stable up to 3 months at 4-12 oC and under transport conditions. However, the stability of IQC sample decreased to only 40 days after the cryotube was opened. The IQC sample for T-CD4 lymphocyte count was developed to be widely used to ensure the quality of CD4 T-lymphocyte test results at laboratories in Vietnam.
