Summary
Memantine hydrochloride is a first-line therapeutic drug approved by the US Food and Drug Administration for the management of moderate to severe Alzheimer’s disease (AD). A number of methods for the synthesis of memantine hydrochloride have been reported. These procedures started from 1,3-dimethyladamantane, 1-bromo-3,5-dimethyladamantane, 1-hydroxy-3,5-dimethyladamantane by as many as using two, three or four reaction steps to produce memantine hydrochloride. In this study, MnCl2 was used as a catalyst in the process to obtain the intermediate compounds 1-acetamido-3,5-dimethyladamantane and 1-formamido-3,5-dimethyladamantane in memantine hydrochloride synthesis reaction. The results showed that MnCl2 revealed a good catalytic activity with high overall yields (93 – 98%) and reducing the ratio molar of the substances involved in the reaction. These intermediate compounds were confirmed by IR, MS, 1H-NMR, 13C-NMR spectrum.
