TÓM TẮT

A high throughput and specific method using ultra performance liquid chromatography tandem mass spectrometry for the determination of the psychotropic agent, quetiapine, in human plasma has been developed. Quetiapine and internal standard (carbamazepine) were extracted from human plasma by protein precipitation technique using acetonitrile. The prepared samples were chromatographed on a C18 column (50 x 2.1 mm, i.d., 1.9 µm) with isocratic elution at a flow rate of 0.4 ml/min using acetonitrile – solution containing 5 mM ammonium formate and 0.05% formic acid at suitable ratio. Detection was carried out by multiple reaction monitoring on a Triple quadrupole mass spectrometer system with an ESI interface. The standard curves were found to be linear in the range 2.0 to 800 ng/ml for quetiapine with mean correlation coefficient greater than 0.998. The intra-day and inter-day precision and accuracy were within 91.5% – 102.1%. The assay was applied to a pharmacokinetic study of quetiapine
in healthy volunteers.
