The HLA-A gene belongs to the superfamily of HLA (Human Leukocyte Antigen) genes, coding the Major histocompatibility complex which plays an important role in human immune system. There have been various reports on the associations of HLA-A polymorphic alleles and the danger of severe adverse drug reactions (ADRs). Genetic testing of HLA genes has been used as pharmacogenetic markers for ADR minimization. This study aimed to establish a simple sequencing protocol for genotyping the HLA-A locus. The protocol with a PCR reaction using 2 primers and a sequencing reaction using 2 primers could identify effectively the genotypes of control samples. This newly established sequencing protocol required less primers, less template DNA than those published previously. The protocol can be applied in Vietnam for genotyping HLA-A gene in personalized medicines.
