Xây dựng quy trình định lượng amygdalin trong đào nhân bằng phương pháp HPLC – PDA

Persicae Semen is a traditional herbal material for manufacturing pharmaceutical products increasing blood circulation, excluding thrombosis. Amygdalin, a cyanogenic glycoside, known as the principally phytochemical component of Persicae Semen could significantly decrease plasma viscosity, prolong the activation time of thromboplastin and reduce fibrinogen content. An HPLC method was developed for determination of amygdalin to assess quality of Persicae Semen. The method was optimized as follows: Column – Phenomenex C18 (250 × 4.6 mm; 5 µm), temperature – 25 oC; Mobile phase – a mixture of methanol and water (21.5 : 78.5), in isocratic mode; Flow rate – 1.0 mL/minute; Injection volume – 20 µL; Detector – photodiode array (210 nm). The method was validated for the suitability of the system, specificity, linearity ( = 19,0924x + 19,9742, R = 0.9999), repeatability (RSD = 1.6%), intermediate precision (RSD = 1.2%), accuracy (recovery rate = 98.4 – 101.6%) and quantification range (9.58 – 38.33 μg/mL). The procedure proved time – saving
(22 minutes for a run) and practically applicable to determination of amygdalin in Persicae Semen.
